Mycoplasma bovis (M. bovis) is one of the important pathogens of cattle, which have been detected from pneumonic lungs of dairy cattle and buffaloes especially calves. M. bovis is main reason for morbidity and mortality in dairy calves because it is overlooked due to its late detection. For the identification of M. bovis in subclinical and clinical samples, determination of acute phase proteins i.e., haptoglobin (Hp) and serum amyloid A (SAA) are important tools for the timely diagnosis of disease. The present study was also performed for detection and phylogenetic analysis of M. bovis induced mortality in bovine calves. The blood, nasal swabs, synovial fluid, lung and pleural fluid samples from bovine calves were collected from slaughterhouses and private farms in Bahawalpur division for AAPs detection and molecular confirmation of M. bovis. Serum was separated form blood samples to determine the concentration of Hp and SAA. The blood plasma obtained from the blood samples was processed to measure stress factors. The lung tissues were processed for culturing on PPLO media to confirm bacteria, while H&E staining method used to checkhistopathological changes in the lungs of effected animals. The DNA was extracted from lung tissues by using DNA extraction kit. The samples were analyzed by PCR using M. bovis-specific primers having a length of 270 bp. The results of present project indicated that level of SAA and Hp were remarkably increased (P<0.05) in M. bovis infected calves in comparison to healthy calves. The oxidative stress markers indicated that nitric oxide and MDA levels in the infected calves increased significantly (P<0.05), while infected bovine claves had considerably lower levels of superoxide dismutase, catalase and glutathione. The PCR results showed that M. bovis was detected in 16(8.29%) isolates, which included 14(7.90%) lung tissues and 2(12.50%) pleural fluid samples. The seasonwise positive samples were 12.12% in winter, 10.34% in spring and 2.89% in summer. The sequences were submitted to NCBI under accession number allotted as PP738878-PP738885. The BLAST was performed for sequence alignment by windows 7.1 software MEGA Muscle 37. The phylogenetic analysis showed the maximum and minimum identity of 97-100% among these geographical obtained strains from local isolates. The reference strain PP738878 had 100% identity with Swiss strain AF003959.1 of M. bovis and other strains showed a minimum identity of 99% with Poland, Switzerland and Egypt. This is the first study of the molecular detection and phylogenetic analysis of M. bovis in bovine calves in Pakistan.
2. Epidemiological Investigation and Development of transdermal agent to Sustainable Parasitic Control in Sheep and Goats
With: ORIC, Islamia University of Bahawalpur (2021-08 - 2023-07)
The main health concern of sheep and goats in rural areas is parasitic infestation which causes decrease in weight gain, poor body condition, decrease in birth weights and difficulty during lambing and kidding. The animals may become vulnerable to further health issues due to parasitic infection which ultimately cause death in animals. Therefore, epidemiological investigation of these parasitic infection is important and need of time in neglected area of Bahawalpur (Cholistan desert). The use of transdermal drug strategy serves dual purposes, it maintains parasitic control in the existence of anthelmintic resistance, and delays the progress of anthelmintic resistance. Therefore, eprinomectin transdermal preparation will be prepared and their efficacy will be addressed through transdermal drug delivery system in sheep and goats. The drug resistance can be controlled because transdermal drug delivery has become a widespread route for the administration of drug in animals to sustainable parasitic control and socio-economic uplift of rural people. Transdermal administration is more convenient to use compared with oral administration and injection administration. It put smaller stress reaction, lower toxic side effects, avoid from the hepatic first-pass metabolism, deliver steady-state plasma level of drug and long-term therapy
